GLYCAN ARRAY SYNTHESIS CORE (D)
Overview
The Glycan Array Synthesis Core (D) produces a library of oligosaccharides for use by Participating Investigators
and other Cores. These compounds represent a diverse range of natural oligosaccharide sequences of
glycoproteins and glycolipids with linker arms compatible for use as monovalent oligosaccharides and
for coupling to biotin, polymers or column supports. Core D has developed a printed mammalian glycan array and a pathogen glycan array that are provided to
Protein-Glycan Interaction Core (H) for the rapid screening of the carbohydrate binding specificity of glycan-binding proteins (GBPs). Both the mammalian glycan array and pathogen glycan array are regularly being expanded by adding synthesized glycans and bacterial polysaccharides onto each array, respectively.
This Core is also producing recombinant proteins including glycosyltransferases and GBPs. Information relating to the production of each material, including novel synthetic procedures, is being deposited in the
Central Database. Available materials are posted on
the website and are distributed to investigators both within and outside of the Consortium upon
approval by the Steering Committee.
Specific Objectives
- Produce a diverse range of natural oligosaccharide sequences of glycoproteins and
glycolipids with linker arms compatible for use as monovalent oligosaccharides and for
coupling to proteins or solid supports
- Work with the Protein-Glycan Interaction Core (H) to create a glycan arrays of mammalian and pathogen oligosaccharides for rapid screening of the binding specificity of GBPs
- Conduct custom synthesis of oligosaccharides by chemoenzymatic or one pot chemical synthesis
methods
- Construct multivalent oligosaccharide probes labeled with biotin or fluorescein or other
labels for detecting cell surface GBPs
- Produce recombinant glycosyltransferases and other enzymes for carbohydrate synthesis
- Produce recombinant GBPs for determination of binding specificity by the
Protein-Glycan Interaction Core (H), crystallography, and
production of antibodies
